What adjustments do I need to make?
500mm, just updated to 2.0.
I've known this has needed adjusting for a while.
What corners and directions do I need to go with this?

I would try right Back Corner Up.
first I would say slow down your rate of z rize while probing and run a larger datapoint mesh.. (9x9 11x11) to get better repeatability. Too fast a bed rize to trip the probe can lead to repeatability/accuracy issues and make sure you do not already have a hieght map turned on
How do I ensure I don't have another heightmap turned on?
HEIGHTMAP from the menu down the left side... if no map is showing you are good to go. If there is one pictured, right above the image of the map click on CLEAR
Depends on your configuration, but usually one is autoloaded. On the main Dashboard , if the position indicators and the little numbers above them are a tad different, then a map is loaded.
