Is it ever acceptable to wear a tie with a chambray shirt?

It can be done but it isn’t as easy to do well as a less casual shirt
The details matter even more than normal if you’re doing something “against the rules”
What are the main concerns with something like this? Like what makes it “against the rules”
Yeah that works. Just make sure the vibe is more business casual than straight up business.
It’s not really against the rules that’s why I put the quotation marks
You just don’t want to put a super formal tie and jacket over a chambray shirt

Tips for doing a chambray shirt with a tie:
Goes better with a casual sports jacket, or suits made with rougher cuts and fabrics
Printed tied and those that can do small knots work best
As most chambray shirts have a small collar, a tie bar can hold it in place

I think the key is a more casual tie like a knit or a wool
Aside from what everyone else said about casual ties and jackets, I would also lean toward a more bizcas shirt. Something with a longer button-down collar would work better than a workshirt with chainstitch runoff. Drake's makes a good chambray shirt meant to be worn with tailoring. Proper Cloth also has a bunch of samples that could work.
I can vouch for the Drake’s chambray - I think it has 3.5” points
